Sumario:The amnion was one of the most important evolutionary novelties in the animal kingdom, allowing independence of water for reproduction and subsequent exploration of terrestrial habitats, and is therefore an important structure to understand evolution. We have studied chicken amniogenesis using ex ovo culture systems and 3D-reconstructions of serially sectioned chicken embryos. We provide evidence for a transient depression of the head in the proamnion, forming a pouch, that positions the extraembryonic membranes dorsal to the head and that is fundamental for the correct formation of the amnion and chorion membranes. When this “sinking” process in the proamnion was blocked, the amnion/chorion did not form, even though the growth of the embryo per se seemed unaffected. Here, we give insight in the role of the proamnion in amniogenesis.
