description Single-cell RNA sequencing (scRNA-seq) is a powerful tool for enumerating the gene expression dynamics at single-cell resolution. Various organs comprising distinct cellular composition and architecture require unique approaches for highly viable single-cell preparation and reliable sequencing results. Here, we describe an optimized protocol for isolating the female reproductive tract (FRT), dissecting different FRT regions, and preparing high-viability single cells from the uterine endocervix and ectocervix to generate a complete molecular cell atlas by scRNA-seq for studying normal physiology and disease.
